Peptide storage and reconstitution: the lab-grade protocol
How to keep lyophilized research peptides stable, what to reconstitute them in, and the mistakes that quietly destroy your reference material.
You paid for reference-grade purity. If you handle the vial wrong after it arrives, you're studying a degradation product, not the compound on the label. This guide covers the boring-but-essential handling steps for lyophilized research peptides.
The one-line summary
Cold. Dry. Buffered. Aliquoted. Skip any of the four and your material degrades faster than you'd expect.
Storage of the sealed vial
Before you reconstitute anything, keep the sealed lyophilized vial at:
- Long-term (>1 month): -20°C, desiccated
- Short-term (<1 month): 4°C, desiccated
- Ship-arrival: the vial should be cold — if it arrived warm, note it and ask the supplier to reship. Lyophilized peptide isn't destroyed by warm shipping instantly, but repeated temperature cycling accelerates degradation.
Desiccation matters. Peptide powders are hygroscopic. Store the vial in a sealed container with silica gel packets, not just on a freezer shelf.
Choosing a reconstitution solvent
The default choice for research peptides is bacteriostatic water (sterile water containing 0.9% benzyl alcohol as a preservative). Use it when:
- The peptide is highly water-soluble
- You need a working solution stable for 2–4 weeks at 4°C
Alternatives exist for less-soluble peptides:

Always add solvent slowly down the side of the vial. Do not vortex vigorously — swirl gently. Peptides can shear or aggregate under mechanical stress.
Concentration calculation
The volume you need depends on your intended stock concentration. Use:
volume (mL) = mass (mg) / target concentration (mg/mL)
Example: to reconstitute a 5 mg vial to 5 mg/mL, add exactly 1.0 mL of bacteriostatic water.
For lower stock concentrations, use more solvent. Most researchers aliquot at 1–5 mg/mL for stability and pipetting convenience.
Aliquoting: the step almost everyone skips
Once reconstituted, immediately aliquot into single-use volumes (50 µL, 100 µL, whatever fits your workflow) and freeze at -80°C. Do not refreeze a thawed aliquot.
Why: every freeze-thaw cycle causes some peptide aggregation and loss of activity. A reconstituted vial that sits at 4°C for weeks and is repeatedly warmed will lose 5–30% activity depending on the peptide — even if it looks fine visually.
Signs of degradation
Look for these before running an assay:
- Cloudiness or precipitate — aggregation. Warm gently (25–37°C for 15 min), swirl. If it persists, the material has degraded.
- Yellow or brown tint in a solution that should be clear — oxidation of methionine or tryptophan residues.
- Weak or absent activity in a validated assay you've run before — usually the first quantitative sign.
If in doubt, re-run identity by mass spec on your working solution. Any research group with an MS line can do this in an afternoon.
Cold-chain shipping — what matters and what doesn't
Legitimate suppliers ship in insulated boxes with gel packs (not dry ice — that's overkill and can crack vials). The vial should arrive cool to the touch. What matters:
- Insulation quality — a real cold-shipping box has 1"+ closed-cell foam, not thin styrofoam
- Gel pack count — 2+ for a small order, more for larger
- Transit time — ideally 24–48 h from ship date
Dry ice is unnecessary for lyophilized peptide in transit. What you need is temperature stability, not extreme cold.
Bottom line
Reference-grade peptide + poor handling = wasted money. A 5-minute protocol on receipt (aliquot, label with date + lot, freeze at -80°C) pays for itself in reproducible results across months of experiments.
